EpiSep CK 8/18 with
DAPI & MCF-7 Cells is used for the direct detection of
cytokeratin 8/18 positive human epithelial cells (i.e. circulating
tumor cells) from human or animal whole blood. It is also
compatible with ascites, pleural effusions, buffy coat, Ficoll
enrichment and cell culture specimens.
Quality Control
Two levels of preserved MCF-7 epithelial cells in 500uL buffered
cell stabilizer are included in this kit to be tested along
with unknown specimens as a quality control measure.
Specificity
The EpiSep Anti-Cytokeratin 8/18 Biotin Conjugate is specific
for human cytokeratins 8 and 18. The immunogens used for this
monoclonal antibody are cytokeratins from the human breast
carcinoma cell line MCF-7. This kit includes DAPI reagent
for identification of nuclei and has been optimized for breast
cancer circulating tumor cell research.
Sensitivity
Direct detection of target cells to levels as low as 1 cell
in up to 1mL of washed whole blood or cell suspensions from
Ficoll, buffy coat, or cell culture.
Recovery
Typical studies using the spiking procedure described here
(link) indicate a recovery of greater than 90%.
